It is possible to use Amazon S3 as a tile storage right now - just upload the tile structrure generated by MapTiler to the S3 bucket. You can use utility like http://s3tools.org/s3cmd or much faster (thanks simultaneous thread upload) utility http://code.google.com/p/s3funnel/
